چکیده

Background and Purpose The therapeutic effects of fluoxetine are believed to be due increasing neuronal plasticity reversing some learning deficits. Nevertheless, a growing amount evidence shows adverse this drug on cognition forms plasticity. Experimental Approach To study the chronic treatment, we combine an automated assessment motivation in mice with investigation central amygdala basolateral amygdala. We use immunohistochemistry visualize types perineuronal nets, along DI staining assess dendritic spine morphology. Gel zymography is used test fluoxetine's impact matrix metalloproteinase-9, enzyme involved synaptic Key Results show that treatment non-stressed increases nets-dependent amygdala, while impairing MMP-9-dependent Further, illustrate how latter contributes anhedonia deficits reward learning. Behavioural impairments accompanied by alterations morphology spines towards immature state, most likely reflecting animals' inability adapt. strengthen link between its influence MMP-9 showing behaviour knockout animals remains unaffected drug. Conclusion Implications Chronic differentially affects various plasticity, possibly explaining opposing brain behaviour. These findings immediate clinical relevance since reported side pose potential threat patients.

Morphine Consumption During Lactation Impairs Short-Term Neuronal Plasticity in Rat Offspring CA1 Neurons

Background: Facing environmental factors during early postnatal life, directly or indirectly via mother-infant relationships, profoundly affects the structure and function of the mammals’ Central Nervous System (CNS). Objectives: This study aimed to evaluate the effect of morphine consumption during the lactation period on short-term synaptic plasticity of the hippocampal Cornu Ammonis 1 (C...

Central amygdala GluA1 facilitates associative learning of opioid reward.

GluA1 subunits of AMPA glutamate receptors are implicated in the synaptic plasticity induced by drugs of abuse for behaviors of drug addiction, but GluA1 roles in emotional learning and memories of drug reward in the development of drug addiction remain unclear. In this study of the central nucleus of the amygdala (CeA), which is critical in emotional learning of drug reward, we investigated ho...

Reward, Motivation, and Reinforcement Learning

There is substantial evidence that dopamine is involved in reward learning and appetitive conditioning. However, the major reinforcement learning-based theoretical models of classical conditioning (crudely, prediction learning) are actually based on rules designed to explain instrumental conditioning (action learning). Extensive anatomical, pharmacological, and psychological data, particularly ...

Optogenetic excitation of central amygdala amplifies and narrows incentive motivation to pursue one reward above another.

Choosing one reward above another is important for achieving adaptive life goals. Yet hijacked into excessive intensity in disorders such as addiction, single-minded pursuit becomes maladaptive. Here, we report that optogenetic channelrhodopsin stimulation of neurons in central nucleus of amygdala (CeA), paired with earning a particular sucrose reward in rats, amplified and narrowed incentive m...
